SEROLOGY
The enzyme-linked immunosorbent assay (ELISA) was designed in the 1960s but came into practical use in 1971. Engvall and Perlman designed an ELISA for use in quantitating IgG. In ELISA, various antigen-antibody combinations are used, always including an enzyme-labelled antigen or antibody, and enzyme activity are measured colorimetrically. The enzyme activity is measured using a substrate that changes colour when modified by the enzyme. The light absorption of the product formed after substrate addition is measured and converted to numeric values.
Haemagglutination inhibition also known as ‘gold standard’ in serology test commonly for Newcastle Disease (ND), Egg Drop Syndrome (EDS) and Avian Influenza (AI). The HI test is based on the principle that the haemagglutinin on the viral envelope can bring about the agglutination of chicken red blood cells (RBC) and this can be inhibited by the specific antibodies.
